Sylabus:

Day 1 Monday: Department of Neuroscience 2.LF UK Plzeňská 130/221, Prague 5 – Motol, building C, 3rd floor, library

Lecture (Vargová): Nonsynaptic transmission as an alternative way of intercellular communication. Extracellular space, extracellular matrix, diffusion parameters in the CNS during physiological states, importance of the nonsynaptic transmission, extrasynaptic “volume” transmission in brain and spinal cord and its mechanism, volume changes in the extracellular space, methods for measuring diffusion parameters in brain and spinal cord.

Practical course (Chmelová) - Manufacturing of microelectrodes used in intra- and extracellular measurements of biopotentials; ion-selective microelectrodes. Fluorescent microscopy and image analysis.

Day 2 Tuesday: Institute of Experimental Medicine ASCR, Vídeňská 1083, Praha 4, campus of the Academy of Science, building LA

Lecture (Vargová): Experimental models of pathological states – ischemia; traumatic lesions; multiple sclerosis; tumors, brain and spinal cord trauma; hydrocephalus; aging; Parkinson‘s disease; Alzheimer‘s disease. The role of extracellular matrix and astrogliosis during pathological states.

Practical course (Vargová): Demonstration of isolated rat spinal cord preparation, tissue slices and preparation of an animal for measurments in vivo (models of pathological states). Diffusion measurements in the CNS .

Day 3 Wednesday: Institute of Experimental Medicine ASCR, Vídeňská 1083, Praha 4, campus of the Academy of Science, building LA

Lecture (Anděrová): Membrane properties of nerve cells and synaptic transmission in the CNS. Membranes of neurons and glial cells; ion channels; patch clamp; receptors; synaptic potentials; gap junctions. Role of glial cells in the CNS during physiological and pathological states. Glial cells types (astrocytes, oligodendrocytes, microglia) and their function; membrane properties; receptors; ion channels; ionic and volume homeostasis; interaction between neurons and glial cells.

Practical course (Kriška): Principles of electrophysiological methods; patch clamp; membrane current measurements; immunohistochemistry; confocal microscopy imaging.

Day 4: Thursday Institute of Experimental Medicine ASCR, Vídeňská 1083, Praha 4, campus of the Academy of Science, building LA

Lecture (Jendelová): Neurogenesis and regeneration of the CNS, cell and gene therapy in the treatment of injured brain and spinal cord, visualization of transplanted cells in vivo using superparamagnetic nanoparticles and magnetic resonance.

Practical session isolation and cultivation of bone marrow stem cells (video)

Day 5, Friday: Institute of Experimental Medicine ASCR, Vídeňská 1083, Praha 4, campus of the Academy of Science, building LA

Lecture (Machová Urdzíková): Preclinical models of spinal cord injury, translation of fundamental research results into clinical medicine, animals used for spinal cord injury models, types of spinal cord injury, simulation of spinal cord injury in animals, testing of motor and sensory function after spinal cord injury.

Practical session (Machová Urdzíková): Models of spinal cord injury (compression lesion and hemisection with hydrogel implantation. Functional tests for spinal cord injury evaluation. (video).
